Here, we propose to use eDNA metabarcoding of water samples as a surveillance tool to detect fish, amphibian, and reptile species of interest to the Texas Military Department (TMD) at Fort Wolters in Mineral Wells, Texas. Specifically, we will use three different primer sets targeting fish (Notropis buccula, N. oxyrhynchus, and N. potteri), amphibians (Pseudacris steckeri and Anaxyrus woodhousii), and snakes (Nerodia harteri, Thamnophis sirtalis, and Thamnophis sirtalis annectens) of interest to TMD. These primer sets will also enable the characterization of the entire fish, amphibian, and reptile community within the study area at Fort Wolters. We will use these data to document diversity and community patterns within and among sites within the study area, as well as across three seasons in order to determine the spatial and temporal variability of these vertebrate assemblages.
