1.1.1: The purpose of this research project is to perform a secondary analysis of hippocampal transcriptomic data from human AD brains. ATRA- and VD-responsive genes were transcriptionally downregulated, accompanied by the upregulation of co-repressors and ROS-sensitive genes Nrf1/2. Therefore, our central hypothesis is that RMAOs are depleted in AD, which is mitigated or prevented by combinatorial RMAO therapy. Using an innovative longitudinal multidisciplinary approach spanning cognitive, biochemical, and multi-omic measures in the next-gen hAβ-KI AD mouse model, we will determine how bidirectional manipulation of RMAO combinatorial therapy alters AD disease trajectories. This is a novel concept compared to AO clinical trials that have failed so far. We predict that combinatorial RMAO therapy will be more effective than the use of generic ROS scavengers.
