Note:
1.1.1: Numerous human diseases are associated with a disruption in redox homeostasis, but addressing molecular mechanisms underlying these events is challenging considering the transient nature of redox changes, such as oxidative modifications and disulfide bond formation. Using a newly identified cytosolic form of peroxiredoxin IV (cPrxIV) as a model protein, we propose to characterize the role of thiol peroxidases in disulfide bond formation in the cytosol, identify cPrxIVinteracting proteins, and investigate the physiological roles of cPrxIV in the reduced cellular compartment. This research has direct implications with regard to mechanisms of redox homeostasis and its disruption in disease.
