Specific Aim 1: To address how TMAO-induced Nlrp3 inflammasome activation contributes to endothelial dysfunction and atherosclerosis in in vivo using Nlrp3-/- mice, endothelium-specific Nlrp3 knockout mice (EC-Nlrp3-/-) and their wild type littermates; Specific Aim 2: To investigate how the non-inflammatory effects of TMAO activated Nlrp3 inflammasomes contributes to impaired endothelium dependent vasodilation, pyroptosis and DAMPs production in primary cultures of CAECs, Nlrp3-/- and Nlrp3+/+ mice; Specific Aim 3: To determine whether TMAO stimulates the formation of MVBs containing NRLP3 inflammasome products in in vitro or in vivo endothelial cells to release exosomes and whether these inflamed exosomes induce endothelial dysfunction and vascular injury.
