SAMs 1.1.1 The goal of the research is to begin the process of bringing resources and know-how to the GCCRI to provide PROTAC technologies to researchers who are interested in targeting “undruggable proteins, such as cancer promoting fusion proteins or their interacting partners. We will build a library of small molecules that bind E3-ligases known to work in the PROTAC paradigm. In addition, we will acquire linkers of varying chemical properties, length and flexibility. Medicinal chemistry approaches for generating linker-E3 binding small molecules will be defined and pursued. Our first candidate “war head” for developing a functional PROTAC will be MI-503, a compound which binds to Menin, a protein involved in acute leukemias. Our second candidate will be the SS18-SSX1 fusion protein involved in synovial sarcomas.
