Note:
we hypothesize that an incoming peptide does not just dock to the fibril tip, but joins a complex located at the tip and that the dynamics of this complex govern the rate of association to the fibril. A corollary of this hypothesis is a novel strategy to suppress fibril growth by stabilizing the intermediate state for peptide incorporation. Notably, the free energy difference between the intermediate complex and the transition state for molecular attachment represents the activation energy for growth so that stabilization of the intermediate state would boost the activation barrier and slow down fibril growth.
